Principle of the Rat ERN1 Endoplasmic Reticulum To Nucleus Signalling 1 ELISA Kit Test

The Rat ERN1 (Endoplasmic Reticulum To Nucleus Signalling 1) ELISA Kit Test is a bioanalytical method that uses the Enzyme-Linked Immunosorbent Assay (ELISA) principle. The test is designed to detect and quantify the levels of ERN1 in a biological sample, such as tissue homogenates and biological fluids. The target antigen (ERN1) in the sample is bound to specific antibodies coated on a microplate. After washing away unbound substances, a biotin-conjugated secondary antibody is added. Then a HRP-enzyme labeled streptavidin is added followed by a color developing substrate. The reaction’s intensity is proportional to the amount of ERN1 in the initial sample.

Importance of the Test

ERN1 is a sensor protein involved in endoplasmic reticulum stress response. It plays a crucial role in homeostatic responses to mitochondrial damage and the unfolded protein response pathway among other processes. Abnormal ERN1 levels have been associated with diseases such as diabetes, Parkinson's disease, and cancer. Therefore, quantifying ERN1 levels can contribute to disease diagnosis, monitoring of disease progression, or effectiveness of treatments.

Advantages of the Test

Procedure to Perform the ELISA

  1. Add 100µL of standard or sample to each well. Incubate 2 hours at 37°C.
  2. Remove the liquid of each well, add 100µL of Detection antibody to each well. Incubate 1 hour at 37°C.
  3. Wash the microplate, add 100µL of Streptavidin-HRP to each well. Incubate 1 hour at 37°C.
  4. Wash the microplate again, add 90µL of Substrate Solution. Incubate 15 minutes at 37°C.
  5. Add 50µL of stop solution to each well to stop the enzyme-substrate reaction.
  6. Measure the absorbance at 450 nm.

Results Analysis

Analyze the measured absorbances using software capable of generating a four-parameter logistic (4-PL) curve fit. The concentration of ERN1 in the sample is determined by comparing the O.D. of the samples to the standard curve.
