The detected OCLN from the sample is captured by the pre-coated antibody and then bound by the biotinylated secondary antibody, and detected by a streptavidin-peroxidase conjugate.

Procedure to perform Rat OCLN Occludin ELISA Kit Test
Add the standard protein or sample to the wells of the ELISA plate.
Add the biotin antibody to all the wells. Then, add the detection enzyme immediately after and incubate.
After the incubation, wash the plate three times with washing buffer.
Add the substrate solution to each well and incubate again.
Finally, add the stop solution, which changes the color from blue to yellow, to each well.
Results Analysis
The results are determined by comparing the O.D. of the samples to the standard curve. Measurements made directly from the samples are then multiplied by the dilution factor to obtain the concentration of Rat OCLN in the original samples.
